Mit den zur Zeit verfügbaren adhärenten Verpackungszellinien ist die Produktion retroviraler Vektoren sehr begrenzt. Das Ziel dieser Arbeit war es, retrovirale Verpackungszellinien zu entwickeln, die in Suspension wachsen und ein 'scale-up' der Vektorproduktion ermöglichen. Es wurde gezeigt, daß Suspensionszellinien fähig sind Retrovirusvektoren transient zu produzieren, doch aufgrund ihrer schlechten Transfizierbarkeit es schwierig ist, eine genügend hohe und stabile Expression der viralen Strukturproteine, sowie des rekombinanten Virusgenoms zu erzielen.
