In the striated muscle, the Z-disc forms the boundary of the sarcomere and contains multiple proteins essential for many aspects of muscle structure and function. At the Z-disc, actin filaments from adjacent sarcomeres are crosslinked by α-actinin, which directly interacts with Zasp PDZ domain proteins. The Zasp PDZ domain family includes the Alp/Enigma family, which is an emerging class of Z-disc-associated proteins, and mutations in many of the family members have been linked to muscle disorders in H. sapiens. Zasp52 is a major D. melanogaster Alp/Enigma family protein, containing a PDZ domain, a Zasp-like motif and four LIM domains. Zasp52 co-localizes with α-actinin to Z-discs, and participates in Z-disc formation. This thesis employs D...
