Opioids are efficacious drugs in the treatment of nociceptive pain but interactions with other drugs can modulate their analgesic efficacy and side effects. Opioids have been shown to activate glial cells in the CNS, which might explain the mechanism of opioid tolerance and opioid-induced hyperalgesia. The aims of the thesis were to study the pharmacological interactions between opioids and diuretics spironolactone, eplerenone, furosemide, and chlorothiazide and anticonvulsant pregabalin in preclinical models. In addition, the effect of chronic morphine treatment on the activation of glial cells in the spinal and supraspinal level was studied.
