Estudo de potenciais marcadores moleculares de suscetibilidade ao câncer de pulmão

Lung cancer is a major public health problem, currently occupying the tenth position among the leading causes of death worldwide and the leading cause of death among cancer. The individual predisposition to developing lung cancer could be associated with genetic polymorphisms related to the inflammatory response, activation mechanisms and detoxification or carcinogens, as well as defects in the mechanisms of the DNA repair. This study aimed to investigate the influence of 13 polymorphisms of the type insertion / deletion in genes of the metabolism and biotransformation (CYP2E1, CYP19A1 and UGT1A1), control genes of the immune system and inflammatory response (IL1A and IL4), genes that regulate control of gene function of the cell cycle and immune system (MDM2 and NFKB1), DNA repair genes (TYMS and XRCC1), regulator of apoptosis gene (CASP 8), regulator of hemostasis gene (PAR1) and control gene cell cycle (TP53) as susceptibility to lung cancer. Polymorphisms were genotyped by a multiplex PCR reaction of patients with a confirmed diagnosis for lung cancer and individuals from the same population without the disease. The genetic ancestry of all individuals were estimated by a panel of ancestry informative markers. A logistic regression analysis controlling for age, gender and smoking was performed to determine the influence of polymorphisms in susceptibility to cancer. No statistically significant differences between the groups with cancer and without cancer were founded. Polymorphisms studied are not associated with susceptibility to lung cancer in the Pará population.O câncer de pulmão é um importante problema de saúde pública, ocupando atualmente a décima posição entre as principais causas de morte no mundo e a principal causa de morte dentre as neoplasias malignas. A predisposição individual ao desenvolvimento de câncer de pulmão pode estar associada a polimorfismos genéticos envolvidos na resposta inflamatória, em mecanismos de ativação ou na detoxificação de carcinógenos, assim como, em defeitos nos mecanismos de identificação e reparo de danos sofridos pelo DNA. O presente estudo teve como objetivo investigar a influência de 13 polimorfismos do tipo inserção/deleção em genes do metabolismo e biotransformação (CYP2E1, CYP19A1 e UGT1A1), genes de controle do sistema imunológico e resposta inflamatória (IL1A e IL4), genes que regulam a função de genes de controle do ciclo celular e do sistema imunológico (MDM2 e NFKB1), genes de reparação do DNA (TYMS e XRCC1), gene regulador da apoptose (CASP 8), gene regulador da hemostasia (PAR1) e gene de controle do ciclo celular (TP53,) quanto a suscetibilidade ao câncer de pulmão. Os polimorfismos foram genotipados por uma reação de PCR multiplex em pacientes com diagnóstico confirmado para câncer de pulmão e em indivíduos da mesma população, sem essa doença. A ancestralidade genética de todos os indivíduos foram estimadas por um painel de marcadores informativos de ancestralidade. Uma análise de regressão logística controlando pelas variáveis idade, gênero e tabagismo foi realizada para determinar a influência dos polimorfismos na susceptibilidade ao câncer. Não foram encontradas diferenças estatisticamente significativas entre os grupos com câncer e sem câncer.
