Abstract: The polyglutamine (polyQ) disorders are diseases of the central nervous system, caused by a CAG triplet expansion in the affected genes, leading to protein aggregation and inclusion formation of the mutant protein in neurons. The size of the expanded CAG repeat correlates with the age at disease onset, yet, up to 50% of the variation in age of onset is determined by other additional factors. Heat shock proteins (HSP) play a central role in protein homeostasis and thus may be crucial in polyQ disease progression. For a selected group of polyQ patients we indeed found that expression of the HSP “DNAJB1” correlated with the age of onset, independent of the length of the CAG repeat expansion.
