Oral Presentation in CongressA genome-wide tagging strategy was followed for the isolation and characterization of novel banana promoters. Embryogenic cell suspensions were transformed with Agrobacterium tumefaciens containing a promoterless, codon-optimized luciferase (luc+) gene next to the right T-DNA border. Around 16,000 transgenic cell colonies were screened two months after transformation at room temperature for baseline luciferase activity and positive colonies were removed. The remaining cell cultures were re-screened in real-time at 26°C followed by a gradual decrease to 8°C.
