PEMISAHAN MINYAK ATSIRI DAUN KEMANGI (Ocimum basilicum ) SECARA KROMATOGRAFI LAPIS TIPIS DAN AKTIVITASNYA TERHADAP Malassezia furfur IN VITRO

Background: Public’s tendency to use natural remedy had been increased recently. Basil’s use as one of natural remedy of traditional plant was still unknown. To know one of its uses, the experiment for Basil’s essential oil isolation by Thin Layer Chromatography and its activity against Malassezia furfur was done. Methods: Essential oil that was used, distillated from Basil’s leaf and separated using Thin Layer Chromatography (TLC). The Silica Gel GF 254 as stationer phase and Chloroform benzene as mobile phase with 1:1 equation and UV light 254 nm as a spot clearing was used. The experimental research with post test only control group design was done to understand Basil’s activity against Malassezia furfur. Five kinds of Essential oil were used in this experiment, consisted of 100%(v/v); 50%(v/v); 25%(v/v); 12.5%(v/v); 6.25%(v/v) concentration and one control group. Sabouraud Dextrose agar (SDA) and olive oil was used as Basil’s medium. Incubation was done for 2 days at 37o temperatures. Evaluation was carried on by seeing fungus’s growth in medium’s surface. Result: Five spots was shown as separation result using TLC which could be observed at 254nm. From microbiology result, there was no fungus’s growth in medium that had been added Basil’s essential oil in all concentration. There was fungus’s growth in control group. After the Mann-Whitney test had been done, was concluded that all essential oil’s concentration, used in experiment against Malassezia fufur was significantly different with p = 0.003. Conclusion: Five components that could be observed were probably belonged in Terpen’s groups. Activity against Malassezia furfur was belonged by Basil’s essential oil. The lowest concentration that was used in experiment still had 100% activity against Malassezia furfur. Key word: Essential oil, Basil, Malassezia furfu
