B-cell receptor signaling pathway The B-cell receptor (BCR) signaling pathway is a central determinant of B-cell fate and func-tion [Wiestner, 2012]. The receptor is a multi-protein structure consisting of a surface transmembrane immunoglobulin (Ig) noncova-lently associated with the Igα (CD79A) and Igβ (CD79B) chains [Dal Porto et al. 2004]. In antigen-dependent signaling, antigenic binding to the BCR causes receptor aggregation and initiation of signal transduction via phospho-rylation of the receptor’s cytoplasmic tyrosine-based activation motifs (ITAMs) by recruited SRC-family kinases, including LYN, FYN, BLK, and LCK [Dal Porto et al. 2004].
