The effect of royal jelly on histone modifications and its role on learning and memory in the honeybee as model organism

Arbeiterbienen und Königinnen haben unterschiedliche Transkriptionsmuster und physiologische Eigenschaften. Da dies durch chromatinmodifizierende Prozesse verursacht werden könnte und die Königinnenentwicklung von einer Fütterung mit Gelée royale (GR) abhängt, habe ich in meiner Dissertation die Frage bearbeitet, ob GR Effekte auf die Chromatinstruktur hat. Mit biochemischen, immunologischen und verhaltensanalytischen Techniken habe ich an der Honigbiene gezeigt, dass GR die Aktivität von Histondeacetylasen (HDACs) in vitro beeinflusst und Histonacetylierungs- und Histonmethylierungsmuster im Gehirn verändert. Da solche Histonmodifikationen auch Lern- und Gedächtnisprozesse beeinflussen, habe ich die Wirkung von GR in Verhaltensexperimenten untersucht. Dabei zeigte sich, dass GR spezifisch und transkriptionsabhängig die Konsolidierung des appetitiven Gedächtnisses nach 24 h und 48 h verbessert. Diese Wirkung kann durch Nikotinamid(NAM) und die 10-Hydroxy-2-decensäure (10-HDA), zwei in GR enthaltenen Substanzen, erklärt werden. Während 10-HDA spezifisch das Gedächtnis nach 24 h verbessert, erhöht NAM spezifisch das Gedächtnis nach 48 h. Sowohl NAM als auch 10-HDA inhibieren HDACs in vitro und führen zu veränderten Histonmodifikationen im Gehirn. Im Vergleich zu GR führen NAM und 10-HDA jedoch zu unterschiedlichen Acetylierungs- und Methylierungsmustern. Deshalb ist anzunehmen, dass es weitere Inhaltsstoffe in GR gibt, die sich auf Enzyme auswirken, die für Histonmodifikationen verantwortlich sind. Zusammenfassend zeigen meine Ergebnisse erstmals, dass GR selbst und einzelne Substanzen in GR auf Enzyme wirken, die an der Modifikation der Chromatinstruktur beteiligt sind und dabei physiologische Prozesse wie die Gedächtnisbildung beeinflussen.Workerbees and queens exhibit different transcription pattern and physiology. Since this is may caused by chromatin modifying processes and due to the fact that queen development is induced by feeding of royal jelly (RJ), I addressed the question whether RJ impacts chromatin structure. Using biochemical, immunological, and behavioural techniques I demonstrated that RJ affects the activity of histon deacetylases (HDACs) in vitro and changes the acetylation and methylation pattern of histones in the brain of honeybees.Since such changes affect learning and memory, I tested the effects of RJ on behaviour. The results show that RJ specifically facilitates consolidation of associative appetitive memory at 24 h and 48 h in a transcription dependent manner. The latter effect can be explained by the additive action of nicotinamide (NAM) and 10-hydroxy-2-decenoic acid (10-HDA), two ingredients of RJ. While 10-HDA specifically improves memory tested at 24 h, NAM enhances memory at 48 h in a transcription dependent manner. Both, NAM and 10-HDA inhibit HDACs in vitro and influence histone modifications in the brain. As compared to RJ however, NAM and 10-HDA have different effects on the histone acetylation and methylation pattern in the brain. This argues for the existence of additonal substances able to act on the machinery that modifies the chromatin structure. Taken together, my results provide first evidence that RJ and distinct substances in RJ act on enzymes involved in the modification of chromatin structure and thus affect physiological processes like memory formation
