Although many facets of biochemical and biophysical research have been reported (14), the function of cellular prion protein (PrPC) remains unclear. A new approach to this issue has emerged from the identification of a homologue, prion protein (PrP)-like protein Doppel (Dpl), with 25 % similarity to two-thirds of the C-termi-nal in PrP (8, 12). The Dpl gene (Prnd) lies down-stream of the PrP gene (Prnp) (12). Interestingly, Dpl is overexpressed in some PrP-null mice to exhibit a late-onset ataxia, besides indicating that ectopic synthesis of Dpl protein is toxic to central nervous system (CNS) neurons (12, 15, 24).
