Modification and evaluation of Abuscreen OnLine assays for drug metabolites

OnLine assays for cannabinoids, barbiturates, opiates, and cocaine from Roche Diagnostics were run on the automatic analyzer COBAS FARA II. The assay programs and the cutoff definition were modified to obtain maximum sensitivity and reagent economy. The limits of detection and curve stability data of the modified assays were compared with the original, as well as with EMIT d.a.u. Cannabinoid 20 assay, and the modified assays were found to be more favorable. Modified OnLine assays appear to be a convenient and cost-effective method for drug screening in urine. Abuscreen OnLine In 1992 Roche Diagnostics (Nutley, NJ) introduced their new assay Abuscreen OnLine using inhibition of latex particle aggregation as a tracer in the Kinetic Interaction of Micro-particles in Solution (KIMS) technique (14-16). In combina-tion with the freely programmable Roche COBAS FARA II rotation analyzer, which can pipette ven very low volumes with the highest precision, this kit seemed to be the basis for developing a cost-effective and high-precision alternative assay to EMIT and fluorescence polarization immunoassay
