Understanding and overcoming resistance to chemotherapy in acute myeloid leukemia For many years, clinical research has tried to identify and overcome the mechanisms of resistance to chemo-therapy in acute myeloid leukemia (AML). Since con-ventional chemotherapy did not allow to improve a CR rate which stayed at 60 % and a DFS which reached 30% at five years at the utmost, with results even far worse in patients aged 60 years or more, the general tendency was to increase dose intensity of antileukemic agents. Allogeneic or autologous bone marrow transplantation is currently proposed during first CR, but the toxic cost of these treatments is still high.
