The autosomal dominant mutations to the amyloid precursor protein (APP), presenilin (PS) 1, and PS2 genes that cause early onset AD have been very informative, leading to the articulation of the amyloid cascade hypothesis.2–4 This hypothesis has been the basis for several disease-modifying therapeutic approaches for AD.
