A New Enzymatic Method for the Estimation of Inorganic Phosphate in Native Sera

A new enzymatic method for the determination of inorganic phosphate (Pi) is described. It utilizes a reaction of the EMBDEN-MEYERHOF pathway in which triosephosphate takes up PI to form glycerate-l,3-diphosphate. It was developed for use with nondeproteinized sera containing high amylase activity. The previously described method based on phosphorolysis of glycogen is not satisfactory in such in-stances and is only half as sensitive as the new method. As the increased sensitivity requires purer reagents, a procedure for removal of contaminating inorganic phosphate and pyrophosphate is given.
