We have constructed a wild buckwheat (Fagopyrum homotropicum) bacterial artificial chromosome (BAC) library. The library contains 24,096 clones stored in 251 96-well microtiter plates. A random sampling of 250 BACs indicated an average insert size of 97 kb. Based on a genome size of 1,080 Mbp, library coverage is 1.9 haploid genome equivalents. Screening the BAC library colony filters with cpDNA sequences showed that contamination of the genomic library with chloroplast clones was 11 %. Library screening with a RFLP probe linked to the S-locus resulted in 2 hits. This library provides for isolation gnen of S-locus
