Frozen sections of kidneys from a one year old non-transgenic C57BL/6 mouse and from three thirteen month old 564Igi mice were stained with fluorescently labeled anti-564 Id (Figure 4 left panels) or anti-mouse IgG2a (Figure 4 middle panels). The C57BL6 kidney sections failed to stain with either antibody (Figure 4). In contrast, anti-Id staining of kidneys from 564Igi mice #1 and #2 resulted in diffuse intense granular (3+) immunofluroescence in glomerular mesangia and some capillary walls. Anti-IgG2a staining of kidneys from mice 1 and 2 stained resulted in 2+ to 3+ immunofluorescence, with the same pattern as seen with the anti-id staining. The kidney of mouse 3 stained more weakly with the anti-Id antibody with trace to 1+ granular deposits in mesangial.glomerular segments. Intense granular (2+) staining in glomerular mesangia was seen with anti-IgG2a staining of the kidney from mouse 3. These results indicated that IgG2a 564 Id+ antibodies were deposited in the kidneys of 564Igi mice. To more definitively document the presence of kidney disease in these mice, kidney sections were examined by electron microscopy (EM) ( Supplementary Figure 2).
