In cancer and various infectious and chronic inflammatory diseases, the quality and quantity of immune cells can be used to predict clinical outcome. For example, the number and location of activated cytotoxic CD8 T cells within human tumors are remarkably accurate prognostic indicators of patient survival. However, immune effector cells including CD8 T cells and NK cells are often functionally paralyzed within the tumor microenvironment due to the expansion of suppressive cell populations such as CD4+CD25+FoxP3+ regulatory T cells, myeloid-derived suppressor cells (MDSCs), tolerogenic dendritic cells or tumor-associated macrophages, and the expression of various suppressive factors including TGF-β, IL-10 and PD-L1.
