Proceedings a/the 9th International Symposium on Buckwheat, Prague 2004 Development and Characterization of RFLP Markers from Buckwheat (Fagopyrum esculentum Moench)

A genomic PstI library from common buckwheat (Fagopyrum esculentum Moench) cv. Sumchanka was constructed and 19 probes were characterized with respect to (a) copy number per genome and (b) similarity of deduced amino acid sequences to known proteins in databases. Approximately 63 % (12) of the probes detected single copy sequences on Southern blots of HindIII-digested buckwheat DNA. Of six probes, three revealed polymorphism between common buckwheat cv. Sumchanka and a line of its wild ancestor, Fagopyrum homotropicum. Thirty percent of the clones show significant similarities to known sequences in protein databases. The use of RFLP probes containing potential genes for genetic mapping will improve the information content of existing meiotic AFLP maps of buckwheat
